Overview
EN ISO 16140-4:2020/A1:2024 is an amendment to ISO 16140-4:2020 published by CEN/ISO. It provides a single‑laboratory protocol for validating a larger test portion size for qualitative microbiological methods in the food chain. The amendment adds a normative Annex H and clarifies procedures for demonstrating that increasing the analysed test portion does not worsen the method’s level of detection (LOD).
Key topics and requirements
- Scope: Validation of a larger measured test portion (volume or mass) used to prepare the initial suspension for qualitative methods. Validation using this protocol applies to the laboratory performing the study and the specific food category tested.
- Annex H (normative): Detailed, mandatory protocol for single‑laboratory validation of larger test portions, including study design, acceptance criteria and interpretation.
- Study design:
- Use a relevant, routinely analysed (and preferably challenging) food item; rationale must be documented.
- Artificially inoculate samples following ISO 16140-2 procedures.
- Prepare at least three contamination levels: negative control, low (target ~theoretical detection ~0.7 cfu/test portion), and higher level.
- Replicates: minimum 1 negative, 20 low-level, 5 higher-level replicates; low-level recovery should be fractional (25–75% positive).
- Statistical evaluation:
- Use the relative level of detection (RLOD) approach: RLOD = LOD_larger / LOD_org.
- Estimate RLOD by fitting a complementary‑log‑log (CLL) model to combined detected/not‑detected data (see ISO 16140-2 Annex D for the model).
- An Excel®-based program for RLOD and LOD calculations is available from the ISO standards site.
- Operational constraints:
- Maintain the validated sample/diluent ratio and incubation conditions when using larger test portions; increases to overcome inhibition are allowed but reductions require validation.
- Pooling rules follow ISO 6887‑1; pooling (test portion) vs pooling (pre‑enrichment) cannot be combined in a single validation-separate studies are needed.
Practical applications and users
- Target users: food microbiology laboratories, method developers, quality control teams, accreditation bodies, and regulatory laboratories that perform qualitative testing for pathogens or indicator organisms.
- Use cases:
- Implementing larger portion sizes to improve detection sensitivity in heterogeneous or low‑contamination food matrices.
- Validating proprietary or alternative qualitative methods before routine use at increased portion sizes.
- Demonstrating method equivalence for accreditation or regulatory acceptance within a single laboratory and food category.
- Post‑validation: If a larger test portion is validated in a multi‑laboratory study per ISO 16140‑2 or ISO 16140‑5, other labs can adopt it after verification per ISO 16140‑3.
